Ultramicroscope: 3D Imaging

Gastvortrag von Univ.-Prof. Hans-Ulrich Dodt (Technische und Medizinische Universität Wien)

„Imaging of cleared biological samples with the Ultramicroscope“

Seminarraum der Fakultät für Chemie, Raum 2124, 1. Stock
Währinger Straße 42, 1090 Wien

Seminar, Workshop, Kurs

New developments in Ultramicroscopy to improve imaging of large cleared biological samples are reported. Objective devices that allow high resolution imaging through 10 mm of clearing solutions are described as well as special optics to create extra thin light sheets. Neuronal networks in whole brains can be visualized. Cleared brains are illuminated with a sheet of light and the optical sections are used for 3D reconstructions. This approach allows one to employ also low power, wide field objectives for imaging of large samples (Figure: e.g. Drosophila melanogaster).
